Peptide Backbone Directed Self‐Assembly of Merocyanine Oligomers into Duplex Structures

چکیده
The pseudopeptide backbone provided by N-(2-aminoethyl)-glycine oligomers with attached nucleobases has been widely utilized in peptide nucleic acids (PNAs) as DNA mimics. Here we demonstrate the suitability of this for formation structurally defined dye stacks. Toward goal a series merocyanine (PMC) connected to were prepared through synthesis. Our concentration-, temperature- and solvent-dependent UV/Vis absorption studies show that under control dipole–dipole interactions, smaller-sized consisting one, two or three dyes self-assemble into duplex structures containing up six chromophores. In contrast, upon further extension oligomer, chosen cannot direct architecture anymore due intramolecular aggregation between dyes. For all aggregate species moderate aggregation-induced emission enhancement is observed.

Nitrogen Backbone Oligomers
We found that nitrogen and hydrogen directly react at room temperature and pressures of ~35 GPa forming chains of single-bonded nitrogen atom with the rest of the bonds terminated with hydrogen atoms - as identified by IR absorption, Raman, X-ray diffraction experiments and theoretical calculations. At releasing pressures below ~10 GPa, the product transforms into hydrazine. Our findings might ...
متن کاملTargeting linear duplex DNA with mixed-base peptide nucleic acid oligomers facilitated by bisPNA openers.
PD-loop complexes formed by bisPNA openers and a mixed-base (mb) oligonucleotide on double-stranded (ds) DNA enable various diagnostic and biotechnological manipulations with linear DNA duplexes [1–5]. So far, the PD-loop formation requires the use of two homopyrimidine bisPNA oligomers to open the DNA duplex for binding an oligonucleotide [6]. This condition imposes certain sequence limitation...
متن کاملAcridine and quindoline oligomers linked through a 4-aminoproline backbone prefer G-quadruplex structures.
BACKGROUND DNA-intercalating drugs are planar molecules with several fused aromatic rings that form stacks between DNA base pairs, reducing the opening and unwinding of the double helix. Recently, interest on intercalating agents has moved in the search for new ligands to G-quadruplex structures. METHODS The DNA binding properties of 4-aminoproline oligomers functionalized with one, two or th...

The accumulation of misfolded proteins as insoluble, fibrillar aggregates is characteristic of several degenerative diseases. Examples include the proteins involved in amyloid diseases such as Alzheimer’s disease (Aβ) (Glenner and Wong 1984), type II diabetes (amylin) (Cooper et al. 1987) and Parkinson’s disease (α-synuclein) (Spillantini et al. 1997), as well as the mammalian prion diseases (P...
متن کاملSecondary structures without backbone: an analysis of backbone mimicry by polar side chains in protein structures.
Backbone mimicry by the formation of closed-loop C7, C10 and C13 (mimics of gamma-, beta- and alpha-turns) conformations through side chain-main chain hydrogen bonds by polar groups is a frequent observation in protein structures. A data set of 250 non-homologous and high-resolution protein crystal structures was used to analyze these conformations for their characteristic features. Seven out o...
